I. Background | |
DLL4(Delta-like ligand 4) 主要表達(dá)于血管內(nèi)皮細(xì)胞,通過 “細(xì)胞-莖細(xì)胞"分 化機(jī)制調(diào)控血管分支:DLL4 激活鄰近細(xì)胞的 Notch1 信號(hào),抑制其分化為細(xì)胞,從而 限制血管過度生長(zhǎng),維持血管網(wǎng)絡(luò)的有序性。在腫瘤微環(huán)境中,DLL4 在異常血管中高表 達(dá),誘導(dǎo)血管結(jié)構(gòu)紊亂(滲漏、低灌注),促進(jìn)缺氧和治療耐藥。此外,DLL4 參與神經(jīng) 發(fā)育及 T 細(xì)胞分化,其基因突變可導(dǎo)致 Adams-Oliver 綜合征等先天性疾病。 | |
